Application of stereology in quantitative pathology - Stereological assessment of the murine intestinal tract
Quantitative pathology is a priority in histopathology labs in order to provide accurate, reliable and reproducible data. Assessment of the functional absorption in mice intestines is often a target in metabolic and microbiota studies. Villi height is one of the parameters frequently assessed in two-dimensional methods; however, these methods are biased and face limitations, which may be overcome by a design-based stereological approach. Stereology is considered the gold standard in quantitative studies, wherein three-dimensional structural knowledge is recovered from 2D histological sections through statistical principles and geometrical measurements. The application of stereology to intestinal studies enables the estimation of various intestinal structural quantities, including intestinal surface area as a qualitative assessment of absorptive epithelia. We propose a method for quantifying intestinal volume and surface area in mouse small and large intestines.
